Dopa oxidase activity in differential diagnosis of amelanotic melanoma tissue.

چکیده
The histopathological diagnosis of melanomas with no demonstrable pigment is difficult. Such tumors are to be differentiated from anaplastic carcinomas, especially of the squamous and adeno types, and from sarcomas showing marked anaplasia or spindle cells. In a previous report from this laboratory (1), a colorimetric method for determination of L-3,4-dihydroxyphenylalanine (dopa) oxidase activity in pigmented tissues was described, and it was suggested that a study of the enzymic oxidation of dopa might be of value in human melanomas. I t is the purpose of this paper to present (a) an improved assay procedure and (b) studies of dopa oxidase activity in melanotic and amelanotic types of human melanoma tissue, other malignant tumors, and non-neoplastic tissues.

CONTROL OF PIGMENT PRODUCTION IN MOUSE MELANOMA CELLS IN VITRO Evocation and Maintenance
A clonally derived amelanotic melanoma cell line repeatedly has been forced to produce pigment by the inhibitor of DNA synthesis, I-/3-D-arabinofuranosylcytosine (ara-C) at sublethal levels . One ara-C-derived melanotic line has been cloned, and has continued to produce pigment for 2 years on normal medium . The inhibitor is most effective when administered to synchronized cells in four pulses ...

متن کاملMSH inhibits growth in a line of amelanotic hamster melanoma cells and induces increases in cyclic AMP levels and tyrosinase activity without inducing melanogenesis.
In Bomirski Ab amelanotic hamster melanoma cells, L-tyrosine and/or L-dopa induce increases in tyrosinase activity as well as synthesis of melanosomes and melanin. L-tyrosine also modifies melanocyte-stimulating hormone (MSH) binding. In this paper we show that in the Bomirski amelanotic melanoma system MSH and agents that raise intracellular cyclic AMP induce dendrite formation, inhibit cell g...
متن کاملUse of antibodies to intermediate filaments in the diagnosis of metastatic amelanotic malignant melanoma.
Immunofluorescent staining of tissue from a lung tumor detected 12 years after excision of a primary malignant melanoma of the skin was negative for prekeratin and positive for vimentin, indicating that the tumor was not epithelial in origin and excluding carcinoma from the differential diagnosis. Complementary conventional staining with hematoxylin-eosin confirmed the melanocytic origin of the...
متن کاملMelanoma detection by enzyme-radioimmunoassay of L-dopa, dopamine, and 3-O-methyldopamine in urine.
This enzyme-radioimmunoassay for the measurement of L-dopa, dopamine, and 3-O-methyldopamine is based on the incubation of urine in the presence of catechol-O-methyltransferase, aromatic-L-amino-acid decarboxylase, and S-adenosylmethionine. The O-methylated dopamine metabolite formed, 3-O-methyldopamine, was characterized by radioimmunoassay. To evaluate the role of L-dopa metabolism in melanom...
